El cultivo del camarón ha generado un gran interés mundial por su creciente demanda en el mercado internacional y su alto valor comercial. La optimización de los parámetros reproductivos en condiciones de cautiverio continúa siendo un desafío para la mayoría de los países productores. En este sentido, la hormona inhibidora de la muda isoforma II (MIH II), descrita para algunos camarones Penaeidae, ha mostrado un papel importante en la maduración ovárica en Metapenaeus ensis y Litopenaeus vannamei, aumentando la expresión de vitelogenina in vitro e in vivo. En este estudio, el ADNc que codifica un péptido maduro de MIH II se aisló mediante técnicas de RT-PCR a partir del pedúnculo ocular de L. vannamei y se clonó en el vector pGEM T Easy. La secuencia nucleotídica deducida consistió en un único ORF de 228 pb que codifica un péptido maduro de 76 aminoácidos. Para su expresión heteróloga en Escherichia coli, el ADNc MIH II se subclonó en el vector de expresión inducible en bacteria pET28a (+) quedando con una cola de seis histidinas hacia el extremo C terminal. La proteína MIH II-His, obtenida en forma de cuerpos de inclusión insolubles, se purificó de forma parcial hasta aproximadamente un 88% de pureza utilizando una cromatografía de afinidad de iones metálicos inmovilizados (IMAC). El efecto de la proteína MIH II-His sobre la maduración y el desove se determinó mediante un ensayo in vivo utilizando hembras de L. vannamei en la fase de intermuda. Se definieron tres grupos experimentales: dos grupos a los que se les suministró una única dosis de 300 y 600 ng/gp, respectivamente, y un grupo control negativo al que se le suministró solución fisiológica de crustáceos. Se registraron los estadios de maduración ovárica y desove durante 72 horas post inyección. Como resultado se observó que en las hembras a las cuales se les suministraron las dosis de 300 y 600 ng/gp ocurrió maduración ovárica en el 8.3% y 33.3%, respectivamente. Las hembras tratadas con la inyección de 300 ng/gp no desovaron y sus gónadas fueron reabsorbidas. Por el contrario, el 100% de las hembras tratadas con la inyección de 600 ng/gp que posteriormente mostraron estadios vitelogénicos III y IV, desovaron. El grupo control negativo no mostró maduración ovárica ni desove. Los resultados preliminares sostienen que la hormona MIH II podría ser un candidato prometedor para inducir el desove en camarones L. vannamei.Shrimp farming has generated worldwide interest due to its growing demand in the international market and its high commercial value. The optimization of reproductive parameters under captive conditions continues to be a challenge for most producing countries. In this sense, the molt- inhibiting hormone isoform II (MIH II), described for some Penaeidae shrimp, has shown an important role in ovarian maturation in Metapenaeus ensis and Litopenaeus vannamei, increasing the expression of vitellogenin in vitro and in vivo. In this study, the cDNA encoding a mature MIH II peptide was isolated by RT-PCR techniques from the eyestalk of L. vannamei and cloned into the pGEM T Easy vector. The deduced nucleotide sequence consisted in a single ORF of 228 bp encoding a mature peptide of 76 amino acids. For its heterologous expression in Escherichia coli, the MIH II cDNA was subcloned into the inducible expression vector in bacteria pET28a (+), leaving a tail of six histidines towards the C-terminus. The MIH II-His protein, obtained in the form of insoluble inclusion bodies, was partially purified to approximately 88% of purity using an immobilized metal ion affinity chromatography (IMAC). The effect of the MIH II-His protein on maturation and spawning was determined by an in vivo assay using L. vannamei females in the intermolt stage. Three experimental groups were defined: two groups administrated with a single dose of 300 and 600 ng/gbw, respectively, and a negative control group that was given a physiological crustacean buffer. The stages of ovarian maturation and spawning were recorded during the following 72 hours post injection. As a result, ovarian maturation occurred in 8.3% and 33.3% of the females administrated with the doses of 300 and 600 ng/gbw, respectively. The females treated with the injection of 300 ng/gbw did not achieved spawn and their gonads were reabsorbed. On the contrary, 100% of the females treated with the injection of 600 ng/gbw that subsequently showed vitellogenic stages III and IV, spawned. The negative control group showed no ovarian maturation or spawning. Preliminary results suggest that the hormone MIH II could be a promising candidate to induce spawning in L. vannamei shrimp.
