    • Marine sponges as a source of new bioactive substances

      Las esponjas marinas (Porifera) han atraído la atención de un gran número de científicos alrededor del mundo. Estos animales y sus simbiontes utilizan mecanismos de defensa química, basados en la producción de compuestos biologicamente activos que utlizan para protegerse de posibles depredadores. Son considerados una mina de oro para los químicos y se han descubierto más de 6000 estructuras novedosas, muchas de ellas con enormes aplicaciones biomédicas, fundamentalmente contra el cáncer, pero también contra diversas bacterias, virus y otras enfermedades. Algunos de los fármacos derivados de esponjas se encuentran disponibles en el mercado, tal es el caso Ara-A (antiviral) y de la Ara-C (antitumoral), pero existen diversos compuestos bioactivos en ensayos clínicos. En Cuba, las investigaciones relacionadas con compuestos obtenidos de esponjas marinas son muy escasas, los descubrimientos iniciales se desarrollaron en el campo farmacológico y solamente en extractos o fracciones. En estudios recientes, se investigaron tres esponjas (Agelas cerebrum, Niphates digitalis y Pandaros acanthifolium), a partir de las cuales se aislaron e identificaron 45 metabolitos, dos de ellos fueron nuevos productos naturales y 30 resultaron metabolitos novedosos, pertenecientes a dos nuevas familias de saponinas esteroidales con características químicas poco comunes y efecto sobre varios parásitos protozoarios, así como contra varias líneas celulares de carcinoma humano.
    • Contribution to the phytochemical study and biological activity of plants of Cuban flora

      Interest in natural products as a source for innovation in drug discovery and agrochemicals is still growing worldwide. Natural products, whose immense diversity has been appreciated for many years, may become in a rich source of novel chemical structures. Our country is a rich source of both biological and chemical diversity which may be useful as a source of novel chemical structures. Even when natural products have been used as medicinal agents for many years in Cuba, their use as agrochemicals are still limited. Thus, the present review focuses on recent advances in the studies on natural products performed by the “Centro de Estudios de Productos Naturales, CEPN” during the past ten years, highlighting on those with potential use as biomedical and agrochemicals. 15 plant species were studied; Agave brittoniana Trel. Subsp. brachypus A Álvarez de Zayas (Agavaceae); Juniperus barbadensis L. var. Lucayana (Britt.) RP Adams (Cupressaceae); Melia azedarach Linn. (Meliaceae); Tectona grandis Linn. f. (Lamiaceae); Lantana camara Linn. (Verbenaceae); Lantana trifolia Cham. (Verbenaceae); Citrus sinensis (Linn.) Osbeck cv Valencia (Ruta- ceae); Maytenus buxifolia (A Rich) Griseb (Celastraceae); Maytenus elaeodendroides Griseb (Celastraceae); Maytenus urquiolae B Mory (Celastraceae); Solanum americanum W. Mill (Solanaceae); Thalassia testudinum Kon. (Hydrocha- ritaceae); Sesbania rostrata Bremek & Oberm (Fabaceae); Pluchea carolinensis G Don (Asteraceae) and Ageratina havanensis (HB & K) RM King & Robinson (Asteraceae). By means of a bio-guided fractionation, isolated structures as well as semi-purified fractions were assayed as antioxidant, antiparasitic, antibiotic, insecticides, antifungal, and skin regenerating and healing effects. The strateg y employed allows us to discover novel secondary metabolites with potential for industrial development as pharmaceuticals and/or agrochemicals. Besides, in addition to the goal of discovering new bioactive agents, the presence of unique bioactive compounds notably contribute to increase the knowledge of plant species in Cuba.
    • Efecto antioxidante y contenido polifenólico de Syringodium filiforme (Cymodoceaceae)

      The marine phanerogam Syringodium filiforme, known as “manatee grass”, is a common species that grows in coastal areas associated to Thalassia testudinum. With the aim to describe some of its possible chemical characteristics, this study was performed with a sample of 1.2kg, collected in March 2009, in Guanabo beach, Havana, Cuba. The sample was dried (less than 12% humidity) and a total extract prepared; other three extracts were prepared with the use of solvents of increasing polarity. The phytochemical screening and analytical determinations of each fraction were undertaken Total polyphenol content was determined using pyrogallol as reference´s standard; chlorophyll a and b and anthocyanin content were also quantified. Total extract and fractions antioxidant activity were evaluated by using the free radical scavenging activity assay with 1,1-Diphenyl-2-Picrylhydrazyl reactive (knowing as DPPH´s method). The phytochemical screening of the different extracts detected the presence of high concentrations of flavonoids, phenols, terpenes, antocyaninns, reducing sugars and alkaloids. The total extract and methanol fraction showed significant free radical scavenging properties, while the petroleum ether fraction showed moderate activity, and the chloroform fraction and the aqueous soluble precipitate (residual salt) obtained didn’t show antioxidant properties against free radicals. The results of this work confirmed the potentialities of this species for biological purposes
    • Acanthifoliosides, minor steroidal saponins from the Caribbean marine sponge Pandaros acanthifolium

      Seven novel steroid glycosides, acanthifoliosides A-F (1-6) and the methyl ester of 6 (7), were isolated from the marine sponge Pandaros acanthifolium as minor components. Acanthifoliosides are characterized by a rare C-15 and C-16 oxidized D ring which was previously found in saponins produced by starfishes. Very uncommon is the presence of additional sugar residues at C-15 or C-16. Their structures were determined on the basis of extensive spectroscopic analyses, including two-dimensional NMR and HRESIMS data. The absolute configurations of the aglycones were assigned by comparison between experimental and TDDFT calculated CD spectra of 1, whereas the absolute configurations of the monosaccharide units were determined by chiral GC analyses of the acid methanolysates. Some of the acanthifoliosides exhibit moderate antiprotozoal activity but to a lesser extent than the most potent pandarosides.
    • Repair of UVB-Damaged Skin by the antioxidant sulphated flavone glycoside Thalassiolin B Isolated from the marine plant Thalassia testudinum Banks ex König

      Daily topical application of the aqueous ethanolic extract of the marine sea grass, Thalassia testudinum, on mice skin exposed to UVB radiation resulted in a dose dependent recovery of the skin macroscopic alterations over a 6-day period. Maximal effect (90%) occurred at a dose of 240 μg/cm2, with no additional effects at higher doses. Bioassay-guided fractionation of the plant extract resulted in the isolation of thalassiolin B (1). Topical application of 1 (240 μg/cm2) markedly reduces skin UVB-induced damage. In addition, thalassiolin B scavenged 2,2-diphenyl-2-picrylhydrazyl radical with an EC50=100 μg/ml. These results suggest that thalassiolin B is responsible for the skin regenerating effects of the crude extract of T. testudinum
    • Parazoanthines A-E: Hydantoin Alkaloids from the Mediterranean Sea Anemone Parazoanthus axinellae

      Five new hydantoin alkaloids, named parazoanthines A-E (1-5), were isolated as the major constituents of the Mediterranean sea anemone Parazoanthus axinellae. Their structural elucidation was achieved through NMR spectroscopic and mass spectrometric analyses. The absolute configuration of the chiral compounds 1 and 4 was determined by comparison between experimental and TDDFT calculated CD spectra. The configuration of the trisubstituted double bond of 2, 3, and 5 was deduced from the 3JH6-C4 coupling constant value. This family of alkaloids represents the first example of natural 3,5- disubstituted hydantoins which do not exhibit a methyl at N-3. All compounds were tested for their natural toxicity (Microtox® assay) and parazoanthine C (3) exhibited the highest natural toxicity. 3 Relatively few chemical studies of zoanthids have been reported so far, despite evidence of their rich natural products chemistry.1 Colonial sea anemones of the genus Parazoanthus have been identified in almost all the oceans and they often have been described as epibionts of marine sponges belonging to the Agelas or Axinella genera. As sponges are known to exude toxic compounds, these zoanthids must have developed adaptative tools to minimize effects of such toxins. Two groups of compounds have been described from Parazoanthus species: fluorescent guanidine alkaloids of the zoanthoxanthin families,2-7 and ecdysteroids.8 As part of an ongoing research program to investigate the chemodiversity of Mediterranean invertebrates,9 specimens of Parazoanthus axinellae were collected near Marseilles (Plane island) as epibionts of the sponge Axinella damicornis. Because the LC–MS analyses of their crude extracts evidenced brominated compounds as major constituents, we decided to undertake the full chemical study of this species from which only zoanthoxanthins have been previously described.2,3 We report herein the isolation and structural characterization of a new class of alkaloids named parazoanthines A-E (1-5) with a rare 3,5-disubstituted hydantoin core.
    • Antiprotozoal steroidal saponins from the marine sponge Pandaros acanthifolium

      The chemical composition of the Caribbean sponge Pandaros acanthifolium was reinvestigated and led to the isolation of 12 new steroidal glycosides, namely, pandarosides E-J (1-6) and their methyl esters (7-12). Their structures were determined on the basis of extensive spectroscopic analyses, including two-dimensional NMR and HRESIMS data. Like the previously isolated pandarosides A-D (13-16), the new compounds 1-12 share an unusual oxidized D-ring and a cis C/D ring junction. The absolute configurations of the aglycones were assigned by interpretation of CD spectra, whereas the absolute configurations of the monosaccharide units were determined by chiral GC analyses of the acid methanolysates. The majority of the metabolites showed in Vitro activity against three or four parasitic protozoa. Particularly active were the compounds 3 (pandaroside G) and its methyl ester (9), which potently inhibited the growth of Trypanosoma brucei rhodesiense (IC50 values 0.78 and 0.038 μM, respectively) and Leishmania donoVani (IC50’s 1.3 and 0.051 μM, respectively).
    • Bromopyrrole alkaloids from the Caribbean sponge Agelas cerebrum

      Bioguided fractionation of Agelas cerebrum crude extract resulted in isolation of four bromopyrrole and four bromopyrrole aminoimidazole alkaloids, identified as 5-bromopyrrole-2-carboxylic acid (1), 4-bromopyrrole-2-carboxylic acid (2), 3,4-bromopyrrole-2-carboxylic acid (3), 4,5-bromopyrrole-2-carboxylic acid (4), oroidin (5), bromoageliferin (6), dibromoageliferin (7) and dibromosceptrin (8) on the basis of spectroscopic data analyses (UV, IR, HRMS, 1D and 2D NMR) and comparison with literature data. This is the first report of compounds 2 and 3 in a marine sponge belonging to the Agelas genus and the first evidence of the presence of 1 from a natural source.
    • Volatile constituents of Thalassia testudinum Banks ex König leaves

      The volatile constituents from the sea grass Thalassia testudinum Banks ex König was analyzed by GC and GC/MS. One hundred and forty-three constituents were identified which constitute more than 95% of the composition. All of them reported for the first time in this species. The most prominent volatile compound was ethyl (Z)-1-propenyl disulfide (31% of the total composition).
    • Polar Alkaloids from the Caribbean Marine Sponge Niphates digitalis

      A method involving flash chromatography, semi-preparative phenylhexyl RP HPLC-DAD-ELSD combined with analytic polar-RP HPLC-DAD, was applied to separate and purify six highly nitrogenated bases and a bicyclic amidine alkaloid, the major components of the marine sponge Niphates digitalis. Their structures were identified as 1,8-diazabicyclo[5.4.0]undec-7- ene (1), deoxycytidine (2), phenylalanine (3), adenosine (4), deoxyguanosine (5), adenine (6) and thymidine (7) on the basis of spectroscopic data analyses. This is the first report of these compounds in a marine sponge belonging to the Niphates genus and the first evidence of the presence of 1 from a natural source.

      The chemical composition of the Caribbean sponge Pandaros sp. was investigated and led to the isolation of seven new steroidal glycosides namely pandarosides A-D (1, 3, 4 and 6) along with the three methyl esters of pandarosides A, C, and D (2, 5 and 7). Their structures were characterized as 3β-[β-glucopyranosyl-(1→2)-β-glucopyranosyloxyuronic acid]-16-hydroxy- 5α,14β-poriferast-16-ene-15,23-dione (1) and its methyl ester (2), 3β-[β- glucopyranosyloxyuronic acid]-16-hydroxy-5α,14β-poriferast-16-ene-15,23-dione (3), 3β-[β- glucopyranosyl-(1→2)-β-glucopyranosyloxyuronic acid]-16-hydroxy-5α,14β-cholest-16-ene- 15,23-dione (4) and its methyl ester (5), 3β-(β-glucopyranosyloxyuronic acid)-16-hydroxy- 5α,14β-cholest-16-ene-15,23-dione (6) and its methyl ester (7) on the basis of detailed spectroscopic analyses, including 2D NMR and HRESI-MS studies. Pandarosides A-D and their methyl esters (1-7) are all characterized by a rare 2-hydroxycyclopentenone D-ring with a 14β configuration.
    • Decoloración de alginato de sodio extraído de las algas pardas marinas del género sargassum con el uso de peróxido de hidrógeno

      Hydrogen peroxide bleaching of sodium alginate from seaweeds oh the Sargassum genus was studied. The influence of H2O2 concentration (percentage of H2O2 on a dry weight alginate basis, w/w) and NaOH/H2O2 ratio (% NaOH/% H2O2, both referred to a dry weight alginate basis, w/w) on the molecular weight, color removal and content of Fe3+ ions of bleached alginate samples was investigated by UV and IR spectroscopies, colorimetric determination of Fe3+ ions and vapor pressure osmometry. Higher yield, purity and molecular weight of alginate were obtained using 3% (or less) of hydrogen peroxide and a NaOH/H2O2 ratio of 1.2 for bleaching.
    • Utilización de la κ-carragenina como soporte de inmovilización de microorganismos marinos degradadores de hidrocarburos.

      Una marea negra provocada por el vertido deliberado o accidental de crudos de petróleo, aceite combustible, fangos o residuos petrolíferos y productos refinados, provoca un gran impacto sobre los ecosistemas marinos y costeros. En este trabajo, se desarrolla un producto compuesto por bacterias marinas inmovilizadas por atrapamiento en k- carragenina semirrefinada y en cuya formulación no se incluyen nutrientes. Los resultados muestran que la variante de inmovilización más ventajosa se obtuvo cuando fue empleada la κ-carragenina al 1% gelificada con una solución de KCL 1.5 M. Mediante análisis de Cromatografía Gaseosa de Alta Resolución y de espectroscopia (FTIR), se demostró la alta capacidad de degradación del bioproducto frente a petróleo del Yacimiento de Pina.
    • The marine plant thalassia testudinum possesses anti-inflammatory and analgesic properties

      The natural marine compounds represent a source of new chemical structures and of pharmacological substances with anti-inflammatory activity that will allow to deep in the knowledge of the inflammatory process and in novel mechanisms of action of therapeutic agents. In this work we carry out the study of a extract of a marine plant present in the Cuban coast, Thalassia testudinum (Tt) with the objective of detecting anti-inflammatory and analgesic effects by carrageenan paw oedema, croton oil mouse ear oedema, phospholipase A2 (PLA2) inhibition, cyclo-oxygenase (COX) inhibition and writhing tests. Three fractions, obtained from this extract, were also evaluated in the assay of croton oilinduced ear oedema, PLA2 inhibition, COX inhibition and writhing test. The extract obtained from the Tt plant demonstrated an inhibitory effect in the carrageenan paw oedema. Also, demonstrated inhibitory effect over PLA2 and COX enzyme. Chloroform and butanolic fractions also inhibited mouse ear oedema assay, and COX enzyme, but only the chloroform fraction inhibited the PLA2 enzyme. The extract, butanolic and aqueous fractions inhibited writhing test. We can conclude that the extract, chloroform and butanol fractions shows antiinflammatory properties and the extract and two of the fractions show analgesic effect.
    • Detección de actividades biológicas en microorganismos marinos aislados de la plataforma cubana

      En este trabajo se evaluaron 126 cepas de bacterias y actinomicetos marinos aislados de diferentes regiones de la plataforma cubana. Los resultados evidencian el potencial biotecnológico de estos microorganismos aislados ya que presentaron actividad contra patógenos considerados como intratables clínicamente y hongos fitopátogenos. Además se detectaron microorganismos potencialmente productores de antitumorales.
    • Efectos farmacológicos de extractos de algas marinas.

      Las algas constituyen una fuente importante de compuestos de interés biomédico. En este estudio se presentan los resultados del pesquisaje realizado a extractos de 7 especies de algas en modelos de conducta exploratoria y convulsiones inducidas por choque eléctrico en ratones machos Balb-c y OF1 respectivamente, de 18-22 g de peso corporal; a dosis entre 40 - 1000 mg/Kg, v.o. (n= 10). Los resultados mostraron que la mayor dosis de los extractos de Dyctiota sp. y Turbinaria sp. disminuyeron significativamente la actividad exploratoria a un 34.9% y 41.6% respectivamente. El extracto de Sargassum sp. disminuyó significativamente la convulsión tónica inducida por el choque eléctrico a un 74.6 %. Estos resultados sugieren la presencia en estos extractos de compuestos con efectos sedante y anticonvulsivante. El extracto de Dyctiota sp. se evaluó en conducta motora y evitación activa en ratas hembras AXC de 200-250 g de peso corporal, así como en conducta exploratoria, laberinto en cruz, natación forzada y convulsiones inducidas por choque eléctrico en ratones machos Balb-c y OF1. Se ensayaron dosis entre 125-1000 mg/kg de peso. Las mayores dosis estudiadas produjeron una disminución de la conducta motora en ratas a 36% mientras que la actividad exploratoria se redujo a 14% en ratones. En el modelo de evitación activa a dosis de 125 y 250 mg/kg del extracto, se observó un enlentecimiento en el aprendizaje. Estos resultados sugieren un posible efecto sedante en este extracto lo que justifica el interés en continuar los estudios con esta especie por su posible interés terapéutico.
    • Aislamiento y caracterización de bacterias heterótrofas procedentes de "Bahía de la Habana".

      En el presente trabajo fueron aisladas, purificadas y caracterizadas bacterias marinas heterótrofas, procedentes de dos estaciones pertenecientes a la "Bahía de La Habana". A partir de un total de 87 aislamientos logrados de diferentes estratos, hubo predominio de individuos Gram positivos. Los mismos, en su totalidad, fueron conservados en cu-ñas de agar cubiertas con aceite mineral, pasando a formar parte de la Colección de Bacterias Marinas del CEBIMAR. Se acometió la identificación de los aislamientos con el empleo de métodos convencionales y también con el sistema API para la taxonomía de bacilos negativos asporógenos (no enterobacterias) y de bacilos esporulados, en-contrándose que varias especies constituyeron nuevos registros para los ecosistemas marinos de Cuba.
    • Actividad biológica en bacterias heterótrofas procedentes de "Bahía de La Habana".

      En el presente trabajo se evaluó la actividad biológica de 87 aislamientos de bacterias heterótrofas procedentes de la "Bahía de La Habana". El espectro de potencialidades mostró que el 29,9% de los mismos presentó actividad antimicrobiana frente a las cepas indicadoras empleadas. Un 63,2% presentó actividad caseinasa, el 44,8% mostró su actividad hemolítica y el 83,4% fue capaz de excretar compuestos con actividad superficial. Respecto a los mecanismos degradativos, el 57,5% de ellas, mostró preferencia por el petróleo como fuente de carbono y el 42,5% por los compuestos organoclorados. Se evidenció la amplia gama de actividades con Bioactividad de bacterias heterótrofas aplicación biotecnológica que poseen las bacterias heterótrofas aisladas de éste ecosistema altamente impactado.
    • Evaluación del efecto genotóxico del D-004 en ratones NMRI empleando la electroforesis alcalina de células individuales en gel (Ensayo cometa)

      El D-004 es un extracto lipídico obtenido del fruto de la palma real (Roystonea regia) que consiste en una mezcla reproducible de ácidos grasos y esteres, en la cual el ácido oleico, el láurico y el palmítico son los más abundantes. Los resultados obtenidos han mostrado que en los modelos experimentales el D-004 presenta una eficacia similar, o ligeramente superior, a los extractos de Saw palmetto en el tratamiento de la Hiperplasia Prostática Benigna. En este trabajo se investigó el efecto genotóxico de la administración oral in vivo del D-004 en ratones NMRI adultos jóvenes de ambos sexos mediante la aplicación de la electroforesis alcalina de células individuales. Se conformaron cuatro grupos experimentales de cuatro ratones, cada uno de ambos sexos, los cuales se trataron con vehículo o D-004 (500, 1 000 y 2 000 mg/kg) por 14 días. Al finalizar el tratamiento, los leucocitos de los ratones de los diferentes grupos fueron embebidos en agarosa, lisados, sometidos a electroforesis y teñidos con nitrato de plata para analizar la ruptura de cadena (RC) y sitios lábiles al alcali (SLA) en el ADN. En correspondencia con otros estudios genotoxicológicos del producto, atendiendo a la ausencia de genotoxicidad reportada para el mismo en los ensayos de Ames y micronúcleos en médula ósea de ratones, los resultados obtenidos en este ensayo no evidenciaron la inducción de RC y SLA.

      Las colecciones microbianas son consideradas como centros de recursos genéticos que custodian dicho germoplasma, entre sus funciones principales se encuentra la conservación de los microorganismos, lo que permite garantizar el suministro de cepas requerido para acometer actividades docentes, investigativas o productivas. En Cuba, tales curadores, se aglutinaron en el Grupo de Colecciones Microbianas, que durante el 2002 se constituyó como Sección de Colecciones Microbianas y otros Materiales Biológicos de la ATAC, alcanzando en la actualidad más de 100 asociados, que participan en reuniones trimestrales, cursos, adiestramientos y talleres, donde son discutidos los resultados del trabajo científico-técnico desarrollado en sus colecciones. Se comentan de modo general su inserción en las distintas esferas de la economía del país, los servicios que prestan, los métodos para la conservación de los microorganismos con que trabajan y otros aspectos de interés.
